If you are asking when ancient Romans turned into medieval nobody can give you an answer which everybody would agree with.If we stay at the most traditional(but only artificial !)date then 476 AD and the deposition of the western Emperor Romulus Augustus is used as a marker between ancient and medieval eras.Problem is that because of its artificiality many more similar markers to devide historical eras exist.Among them:

-Emperor Diocletianus establishment of the "Dominate" form of rule since 284 AD
-The Empire is permanently devided between two Emperors at west and at the east to be never again united under one ruler.
-Death of the Emperor Justinian I(who is often seen as the last "Latin" Emperor)
-Death of Emperor maurice in 602 AD
-Emperor Heraclius reforms starting from 610 AD
-Iconoclasm in the Empire starting from 730(seen as an end of the antiquity culture)
